大蒜辣素對db/db糖尿病小鼠心肌細胞凋亡及Pink1

目的: 觀察大蒜辣素對db/db糖尿病小鼠心肌細胞凋亡及Pink1/Parkin信號通路的影響。方法: 在db/db糖尿病小鼠模型的基礎上,隨機分為糖尿病模型組及大蒜辣素組,同時設正常對照組。連續干預21 d后處死剪取左心室組織。采用TUNEL染色法分析各組心肌細胞的凋亡水平;采用Western blot及qRT-PCR檢測Pink1、Parkin及Mfn1的表達情況。結果: 糖尿病模型組心肌細胞凋亡明顯增多,大蒜辣素組心肌細胞凋亡改善明顯;糖尿病模型組Parkin表達明顯降低,大蒜辣素組表達明顯增多,而Pink1的表達水平各處理組無明顯差異。糖尿病模型組心肌細胞Mfn1表達明顯增高;大蒜辣素組與糖尿病模型組相比,Mfn1表達明顯降低。結論: 大蒜辣素可有效抑制糖尿病心肌細胞凋亡,并通過激活Pink1/Parkin信號通路以保護心肌。
